The Cornell group is investigating pathological mechanisms underlying common and rare genetic variants associated with increased risk for orofacial clefting. Current projects focus on gene regulatory network governing differentiation of periderm and neural crest. A lab member is need to conduct functional tests of human variants that potentially cause orofacial clefting. Exome analyses of orofacial clefting patients have detected coding variants in genes known to be required for embryonic development. To test if these variants disrupt the function of the encoded protein, we inject mRNA corresponding to the reference variant, or the patient variant, into zebrafish embryos mutant for the orthologous gene and assess rescue of the mutant phenotype. Other methods deployed in the lab include Cut-and-run, RNA-seq, ATAC-seq, nNanostring profiling, genome engineering, and in vivo reporter assays.
